The myxovirus resistance gene (Mx1) has a broad spectrum of antiviral activities. It is therefore an interesting candidate gene to improve disease resistance in farm animals. In this study, we report the use of somatic cell nuclear transfer (SCNT) purton vets to produce transgenic pigs over-expressing the Mx1 gene. These transgenic pigs express approximately 15 25 times more Mx1 mRNA than non-transgenic pigs, and the protein level of Mx1 was also markedly enhanced. We challenged fibroblast cells isolated from the ear skin of transgenic and control pigs with influenza A virus and classical swine fever virus (CFSV). Indirect immunofluorescence assay (IFA) revealed a profound decrease of influenza A proliferation in Mx1 transgenic cells. Growth kinetics showed an approximately 10-fold reduction of viral copies in the transgenic cells compared to non-transgenic controls. Additionally, we found that the Mx1 transgenic cells were more resistant to CSFV infection in comparison to non-transgenic cells. These results demonstrate that the Mx1 transgene can protect against viral infection purton vets in cells of transgenic pigs and indicate that the Mx1 transgene can be harnessed to develop disease-resistant pigs. Type I interferons (IFNs) are important mediators of the innate immune responses and are crucial for limiting the early replication and spread of viruses [ 1 ]. An important downstream effector of type I IFNs is the myxovirus resistance gene (Mx1 in mice and pigs, MxA in humans). The expression of Mx1 is strongly induced by IFN-α/β,double-stranded RNA or viral infections purton vets [ 2 , 3 ]. The Mx1 protein can protect numerous purton vets hosts by preventing the growth of a wide variety of viruses both in vitro and in vivo including orthomyxoviruses [ 4 - 6 ], bunyaviruses [ 7 , 8 ], rhabdoviruses [ 9 ], paramyxoviruses [ 10 ] and hantaviruses [ 11 , 12 ]. Pigs are susceptible to swine, human and avian influenza viruses. As alternate hosts, pigs are believed to be mixing vessels by enabling the genetic rearrangement of influenza purton vets viruses and thereby purton vets amplify their genetic variation. As a consequence, the probability of influenza pandemics threatening humans is increased [ 22 ]. A recent report revealed that the circulating pandemic H1N1/2009 influenza A virus originated in pigs and contained genetic contributions of human, avian and swine influenza viruses [ 23 ]. Therefore, influenza resistant pigs would not only benefit the livestock industry but also global public health by eliminating a critical host so that new variants of pandemic influenza strains can no longer emerge. Mx1 is an interesting candidate gene to engineer pigs with enhanced viral resistance. However, it may be associated with the risk of transmission of infectious porcine liphook equine pathogens. Porcine endogenous retroviruses (PERVs) are of particular concern because they have been shown to infect human cells in vitro. To date, researches on the molecular characteristics and potential pathogenicity of PERV are still tenuous. In this report, an infectious replication competent clone of PERV from Wuzhishan pigs (WZSPs) in China was generated and characterized. This infectious clone will contribute to studies on PERV virology and control of PERV in xenotransplantation using Chinese miniature pigs. Methods
The proviral DNA of PERV from WZSPs was amplified in two overlapping halves. Then the two fragments were isolated, subcloned and fused to generate pBluescriptαSK + -WZS-PERV recombinant clones. Screened with RT-PCR, a molecular clone of PERV designated as WZS-PERV(2) was selected. Its infectivity and replication competency were characterized in HEK293 cells by PCR, real-time fluorescent quantitative liphook equine RT-PCR, western blot, indirect immunofluorescence assay as well as sequence analysis. liphook equine Results
The ability of WZS-PERV(2) to infect human cells and produce infectious virions were shown after transfection of the clone into HEK293 cells and infection of PERV derived from this recombinant clone. The expression of Gag proteins were detected in HEK293 cells infected with the virus derived from the clone by the indirect immunofluorescence assay and western liphook equine blot. The results of sequences analysis and comparison combined with the PCR based genotyping result demonstrated that the WZS-PERV(2) belonged to PERV-A subgroup. Compared with a previous proviral DNA clone of PERV (PERV-WZSP), G to A hypermutation occurred in the env gene of WZS-PERV(2) was found, whereas APOBEC proteins have the potential to inhibit the replication of a variety of retroviruses through a cDNA cytosine deamination mechanism, liphook equine so we presumed these G to A hypermutation might be the contribution of porcine APOBEC3F. Pigs are the most suitable candidate because of the anatomical and physiological similarities shared with humans as well as ethical concerns [ 1 - 3 ]. However, it may be associated with the risk of transmission of infectious porcine pathogens [ 4 - 7 ]. Most transmissible porcine microorganisms to human recipients can be precluded by specific-pathogen-free (SPF) breeding, but this is not possible in the case of porcine endogenous retroviruseses (PERVs), liphook equine which are integrated into the genome of all pigs and can stably inherit in the germ line.
PERVs belong to C-type retroviruses and are classified to three subtypes, PERV-A, PERV-B and PERV-C, according to the env sequences [ 8 ]. PERV-A and PERV-B are characterized with polytropism and can productively infect a wide spectrum of human and other mammalian cells. In contrast, PERV-C is ecotropic viruses and can only infect pig cell lines. Additionally, PERV A/C recombinants have recently been reported. Pluripotent stem cells have the potential streatham hill vets to become any cell in the adult body, including neurons and glia. Avian stem cells could be used to study questions, like vocal learning, that would be difficult to examine with traditional mouse models. Induced pluripotent stem cells (iPSCs) are differentiated cells that have been reprogrammed to a pluripotent stem cell state, usually using inducing genes or other molecules. We recently succeeded in generating avian iPSC-like cells using mammalian genes, overcoming a limitation in the generation and use of iPSCs in nonmammalian species (Rosselló et al., 2013). However, there were no established optimal cell culture conditions for avian iPSCs to establish long-term cell lines and thus to study neuronal differentiation in vitro . Here we present an efficient method of maintaining chicken iPSC-like cells and for differentiating them into action potential generating neurons. 1. Introduction
Pluripotent stem cells are undifferentiated cells that have the potential to become any cell type in the adult body. They have two main hallmarks. First, unlike differentiated somatic cells, stem cells have the ability to self-renew and proliferate. Second, they have the ability to differentiate into somatic cells with drastically different properties. streatham hill vets There are currently two main methods that can be used to derive or create stem cells in vitro . The first is to isolate embryonic stem cells (ESC) from early embryos. The second is to induce pluripotent stem cells (iPSCs) from somatic cells such as fibroblasts, first demonstrated possible with a combination of four transcription factors: Oct4, Sox2, Klf4, and c-myc [ 1 ].
Though both ESCs and iPSCs can differentiate into somatic cells, there are differences between the two types of stem cells that still elicit concerns of whether iPSCs could fully recapitulate the characteristics of pluripotent streatham hill vets ES cells. These include iPSCs genetic alterations streatham hill vets from integration of the viral vector streatham hill vets containing the reprogramming transcription factors into host genes [ 2 ] and epigenetic modifications such as DNA methylation and histone activation [ 3 ]. Birds are important organisms for studying development, including neural development [ 7 ]. One of the advantages streatham hill vets to using chicken as a model organism is the ability to monitor its embryo development in ovo . However, a limitation of studies with birds is that there are very few cell lines available for studying cell function. Thus, one of our goals has been to create avian stem cells that could be used to study neuronal function. Towards streatham hill vets this goal, we recently were successful at using the mammalian homologs of the iPSC inducing transcription factors to induce partial iPSC-like cells across the animal kingdom, in birds, fish, and Drosophila [ 8 ]. We were able to show in vivo incorporation and differentiation streatham hill vets of the cells in birds and fish but were not able to obtain stable cell lines that survived many passages. Here we developed novel media conditions that allowed us to maintain these same avian cells well past 20 passages, and we were able to differentiate them into neurons in vitro . 2. Materials and Methods
